Method Development
We are constantly working on improving the advanced fluorescence microscopy methods we are using in our lab, many of which we developed ourselves. We developed and implemented a novel particle tracking method based on movable confocal pinholes and combined with z-Stacking of widefield images. We are permanently optimizing our multicolor ALEX-TIRFM setup with regard to accuracy and imaging speed, enabling us to detect smallest variations in FRET efficiencies of molecule complexes. In confocal microscopy, we introduced Pulsed Interleaved Excitation (PIE), a method to either remove spectral crosstalk or calculate FRET efficiencies in Laser scanning images or FCS measurements.
